Inflammatory Screening –Chemotaxis Assay

Cell migration is a fundamental function of many cellular processes including immune response and inflammation. Leukocytes normally circulate the body in a resting state until the come into contact with chemicals known as chemoattractants. They rapidly respond by adhering to and migrating through the endothelial layer in the vasculature towards the site of inflammation. This directional movement of cells along a concentration gradient of a chemical in their environment is referred to as chemotaxis.

Chemotaxis appears to play an important role in leukocyte recruitment and therefore constitutes a target for anti-inflammatory drug discovery, whereby the goal is to develop methods to control inflammation by modulating or blocking leukocyte chemotaxis to the site of inflammation.

Assay Principles

GlycoMar’s inflammatory cell chemotaxis assay determines the migration of labelled human neutrophils towards a chemoattractant molecule. The assay is based on the boydan chamber principle in which labelled Neutrophils are placed in an upper chamber, separated from the lower chamber by a porous membrane. The lower chamber contains media and/or a chemoattractant. Test samples can be added to the top chamber along with the Neutrophils or to the bottom chamber in place of the chemoattractant. This assay is very flexible and can be run with or without a monolayer of Human umbilical vein endothelial cells (HUVECs) and with a variety of chemoattractant compounds.

Results

Cell migration is calculated as percentage of cells migrated relative to total cells. Results are reported, summarized graphically, in a format and timescale agreed with the client. The assay is suitable for the performance of dose-response studies.

Sample Submission

Samples can be submitted to GlycoMar by prior arrangement, in a number of formats.

Pricing is dependant on sample numbers submitted and turnaround required. Currently the assay is available in a 24-well transwell format offering low – medium throughput.

Standard turnaround is 4 weeks from receipt, but faster turnaround is possible on request.
